Objective: To identify a new strategy for postoperative pain management, this study investigated the analgesic effects of allopregnanolone (Allo) in an incisional pain model, and its effects on the activities of the primary afferent fibers at the dorsal horn.
Summary: Systemic administration of Allopregnanolone inhibited mechanical allodynia and activities of the primary afferent fibers at the dorsal horn in a rat postoperative pain model. Allopregnanolone was proposed as a candidate for postoperative pain management.
Usage: Fluorescence immunohistochemistry: Six sections per animal were labeled with primary antiserum, rabbit anti-NK-1R (1:3000).

Summary: P2YR’s are metabotropic purinergic receptors found in some parts of the CNS. A subtype of this receptor excites rhythm generating networks in the preBötzinger complex. In order to better understand the role of these receptors in modulation of motor output the authors used brainstem-spinal cord and medullary slice preparations from neonatal rats to investigate P2Y1R signaling on specific neurons that innervate diaphragm and airway muscles. Anti-NK1r (Cat. #AB-N33AP) at a 1:1000 dilution was used during the immunohistochemistry. The data suggest that loss of purinergic modulation contributes to motoneuron excitability.

Summary: In this work the authors utilized three different preclinical pain models to examine the effects of intrathecal administration of the DOR agonist SNC80. One strategy was to assess NK-1 receptor (NK1r) internalization using anti-NK1r (Cat. #AB-N33AP) in IHC at a 1:3000 dilution on cryosections. The data indicate that the transmitter release from small peptidergic afferents is an effect mediated by DOR in the spinal cord.

Summary: Pharmacological experiments have suggested that sighs and normal respiration are highly dependent on calcium currents carried by P/Q channels. Using transgenic mice missing the Cav2.1 pore-forming α1A subunit the authors demonstrate that loss of P/Q-type calcium channels results in compromised breathing, sighing, neuromodulation, and leads to early death. A neurokinin-1 receptor antibody (Cat. #AB-N33AP) at a 1:500 dilution was used for immunohistochemistry in this work.

Summary: In this work the authors administered 100 ng SSP-SAP (Cat. #IT-11) into the intrathecal space of rats (saporin, Cat. #PR-01, was used as a control). Lesioned animals displayed decreased carrageenan-induced mechanical allodynia, and carrageenan-induced phosphorylation of Akt was blocked throughout the spinal cord gray matter.
