Objective: Usage of novel fluorescent DNA-based probe technology to map activities of endogenous NOS3 in live cells provides a platform to discover selective regulators of NOS3 in PM and TGN.
Summary: Targeting of plasma membrane (PM) and trans-Golgi network (TGN) shows that NOS3 is tenfold less active at the Golgi but its activity is essential for the structural integrity of the Golgi.

Objective: To investigate whether there is a molecular mechanism underlying the regulation of LTBP1-TGF-b signaling by the BH4-biosynthesis pathway.
Summary: GTP cyclohydrolase I (GTPCH), 6-pyruvoyltetrahydropterin synthase (PTPS), and sepiapterin reductase (SR) are sequentially responsible for de novo synthesis of tetrahydrobiopterin (BH4), a known cofactor for nitric oxide synthase (NOS). PTPS metabolic activity is required for colorectal tumor cell growth under hypoxia.

Objective: To evaluate the effect of water pipe smoke on oxidative stress levels in buccal cells, using MAWI i-SWAB tubes for the detection of different protein oxidation biomarkers (S-glutathionylation, S-nitrosylation and tyrosine nitration). Furthermore, these tubes were used for the revelation of iNOS and NOX2.
Summary: Results confirmed the findings about the ability of these tubes to detect S-nitrosylation, nitrotyrosine, as well as S-glutathionylation. Results showed also the presence of immature NOX2 in human buccal cells equally.

Objective: To examine central mechanisms for the (±)-epibatidine-induced responses, focusing on brain NOS and NO-mediated mechanisms, soluble GC (sGC) and protein S-nitrosylation (a posttranslational modification of protein cysteine thiol groups), in urethane-anaesthetized (1.0 g·kg1, i.p.) male Wistar rats.
Summary: Stimulation of brain nAChRs can induce elevation of plasma catecholamines through brain iNOS-derived NO-mediated protein Snitrosylation in rats. Therefore, brain nAChRs (at least α4β2 subtype) and NO might be useful targets for alleviation of catecholamines overflow induced by smoking.

Objective: To investigate the oxidative profile of iSWAB-Mawi protein tubes by examining their ability to detect the protein oxidation biomarkers: nitrotyrosine and S-nitrosocysteine in buccal cells.
Summary: Results showed the presence of S-nitrosocysteine and nitrotyrosine in buccal cells reflecting the ability of iSWAB-Mawi protein tubes to detect those protein oxidation biomarkers.

Objective: To investigate the molecular mechanisms underlying chitosan nanoparticles containing S-nitrosomercaptosuccinic acid (S-nitroso-MSA-CS) induced cytotoxicity in melanoma cells.
Summary: Melanoma cells were more sensitive to cell death than normal melanocytes. S-Nitroso-MSA-CS-induced cytotoxicity exhibited features of caspase-dependent apoptosis, and it was associated with oxidative stress, characterized by increased mitochondrial superoxide production and oxidation of protein thiol groups.
