Targeting Tools: Targeting B-Cell Leukemias and Lymphomas

Anti-CD22 (UV22-2) Mouse Monoclonal

This IgG1k antibody (Cat. #AB-36) recognizes the group A epitope of the human B-cell antigen, CD22. Clone UV22-2 reacts exclusively with cells of human B-cell lineage.

The fluorescent conjugate mouse IgG-FITC (Cat. #FL-07) is available as a research tool to use with this and other mouse monoclonal antibodies.

Daudi cells, a Burkitt lymphoma cell line, were used in flow cytometry with a mouse monoclonal antibody to CD22 (Clone UV22-2). Cells were treated with 0.5 μg per million cells of Anti- CD22 mouse monoclonal and subsequently with anti-murine IgG- FITC (Cat. #FL-07). A 99% shift is seen as compared to the secondary alone.

Anti-CD22-SAP Immunotoxin

This immunotoxin (Cat. #IT-37) will specifically eliminate cells that possess the group A epitope of the human B-cell antigen, CD22. Cells that do not express CD22 will be unaffected.

This immunotoxin is also available in a kit (Cat. #KIT-37) that includes equal amounts of immunotoxin, unconjugated saporin (Cat. #PR-01), unconjugated antibody (Cat. #AB-36), and a control immunotoxin, Mouse IgG-SAP (Cat. #IT-18).

Daudi cells, a Burkitt lymphoma cell line, were plated at 3000 cells per well and were allowed to acclimate. Samples were added at the indicated concentrations and cells were incubated for 72 hours. MTS (Promega) was added and, after color development, wells were read with a Molecular Dynamics SpectraMax 340. ED50’s of each compound are color-coded. Data analysis is by PRISM (GraphPad).
