Objective: To elucidate the mechanism through which EMA300, a small molecule antagonist of the angiotensin II type 2 receptor (AT2R) with >1,000-fold selectivity over the angiotensin II type 1 receptor, produces analgesia in a rodent model of neuropathic pain.

Summary: Augmented angiotensin II/AT2R signaling in the DRGs of CCI rats is attenuated by EMA300 to block p38 MAPK and p44/p42 MAPK activation, a mechanism with clinical validity for alleviating neuropathic pain.

Objective: To immunohistochemically differentiate the angiotensin receptor subtypes: AT-1ARa, AT-1ARb, and AT-2R.

Summary: Antibodies that can differentiate the three different angiotensin II receptor subtypes in the rat were used to immunohistochemically label angiotensin II receptor subtype-like immunoreactivity in the rat adrenal, pituitary, and brain. The pattern of staining corroborates mRNA, radioligand binding, and functional studies of adrenal and anterior pituitary angiotensin receptors.

Usage: Western blot (1:500), immunofluorescence (1:500), immunocytochemistry (AB-N25AP and AB-N26AP at 1:500, AB-N28AP at 1:2000).

Objective: To determine interactive sites in the sensory vagal complex of rat brain using dual immunolabeling of antisera recognizing the AT1-AR and Ang II.

Summary: In the rat sensory vagal complex, AT-1A receptors are strategically positioned for involvement in modulation of the postsynaptic excitability and intracrine hormone-like effects of Ang II. In addition, these receptors have distributions consistent with diverse roles in regulation of transmitter release, regional blood flow and/or vascular permeability.
