Objective: To evaluate the effect of extensive water-soluble chitosan (EWSC) in Allergic rhinitis (AR).
Summary: The results suggest that NGF- and TH2-related cytokines created a positive feedback loop amplifying allergic inflammation in the upper-airway epithelial cells, and EWSC attenuated allergic inflammation and epithelial cell damage, by inhibiting NGF biosynthesis during allergic TH2 immune responses.

Objective: To investigate whether Sox2 is required during the process of melanomagenesis, melanoma growth and metastasis and in the acquisition of resistance to BRAF inhibitors (BRAFi) treatments.
Summary: The findings exclude an oncogenic function for Sox2 during melanoma development and do not support a role for this transcription factor in the acquisition of resistance to BRAFi treatments.

Objective: To characterize the effects of the neurotrophins nerve growth factor (NGF), brain derived neurotrophic factor (BDNF) and neurotrophin 3 (NT-3) on the sprouting rate of sympathetic and parasympathetic neurites from the female mouse ganglion.
Summary: All three factors (BDNF, NT-3 and NGF) stimulated neurite outgrowth of both sympathetic and parasympathetic neurites although BDNF and NT-3 had a higher stimulatory effect on parasympathetic ganglion cells. Active forms of BDNF and NT-3 were detected in urinary bladder tissue, whereas tissue from the diaphragm expressed NGF.

Objective: To show that Sox10+ adult stem cells contribute to both encapsulation and microvessel formation.
Summary: This study provides a novel mechanism that Sox10+ adult stem cells in the stroma of subcutaneous loose connective tissues are a common precursor of fibroblasts/myofibroblasts and perivascular cells. These adult stem cells can first differentiate into fibroblast-like cells at early stages of biomaterials implantation, and then into myofibroblasts promoting encapsulation/fibrosis, or perivascular cells supporting microvessels.

Objective: To compare survival and migration of murine boundary cap neural crest stem cells (bNCSCs) and embryonic stem cells (ESCs)-derived, pre-differentiated neuron precursors after their implantation acutely at the junction between avulsed dorsal roots L3-L6 and the spinal cord.
Summary: The data show that both stem cell types successfully survived implantation to the acutely injured spinal cord and maintained their differentiation and migration potential. The data suggest that, depending on the source of neural stem cells, they can play different beneficial roles for recovery after dorsal root avulsion.

Summary: Immunochemistry analysis with p75 analysis confirmed that the cells exhibited a Schwann cell phenotype, suggesting that electrospun polycaprolactone/chitosan scaffolds would be good candidates for peripheral nerve tissue engineering.

Summary: The ability of neonatal mouse cochlear supporting cells to divide and differentiate into hair cells is very limited and declines in the first two weeks after birth. This decline is associated with the morphological and functional maturation of the organ of Corti prior to the onset of hearing, however little is known of the molecular changes that underlie these events. The authors attempt to identify these changes using RNA-seq to generate transcriptional profiles of purified cochlear supporting cells and found significant changes in gene expression related to regulation of proliferation, differentiation of inner ear components and the maturation of the organ of Corti. The authors also examined the regenerative potential of supporting cells in production of hair cells in response to a blockade of the Notch signaling pathway at the time of birth, but a complete lack of response just a few days later. Analysis included IHC on frozen sections of paraformaldehyde-fixed temporal bones of LfngEGFP mice. Anti-NGFr (mup75) (Cat. #AB-N01AP) was used at a 1:200 dilution. The results offer first molecular insights into the failure of hair cell regeneration in the mammalian cochlea.

Objective: To investigate the therapeutic effect of induced pluripotent stem cell-derived neural crest stem cells (iPSC-NCSCs) and mesenchymal progenitor cells (MPCs) on peripheral nerve regeneration.
Summary: Transplantation of NCSCs has better outcomes of motor nerve recovery and muscle reinnervation by Schwann cell differentiation in vivo and paracrine signaling, whereas transplantation of MPCs fails to promote functional nerve regeneration.

Objective: This study aimed to investigate the beneficial effects of Cheonggukjang (CGK) manufactured by mixed culture of Bacillus subtilis MC31 and Lactobacillus sakei 383 on neurotoxic damages.
Summary: The short- and long-term memory loss induced by trimethyltin (TMT) treatment was significantly improved in the CGK-pretreated group in a dose-dependent manner. A dose-dependent increase in nerve growth factor (NGF) concentration, activation of the NGF receptor signaling pathway including the TrkA high affinity receptor and p75 NTR low affinity receptor was measured in all TMT/CGK-treated groups.

Summary: Huntington’s disease (HD) is a dominantly inherited neurodegenerative disorder. It’s characterized by a combination of motor, cognitive, and psychiatric features. Striatal spiny neurons are dependent on brain-derived neurotropic factor for proper function and survival. Studies suggest both the receptors for BDNF, TrkB and the p75 neurotrophin receptor (p75), are improperly regulated in the striata of HD patients and mouse models. The authors investigated the role of p75 in the Q175 knock-in mouse model of HD be examining levels of activation of downstream signaling molecules to determine if p75 represents a promising therapeutic target. Anti-NGFr (mup75) (Cat. #AB-N01AP) was used at a 1:2000 dilution in immunoblotting. The data suggest that p75 signaling plays an early role in augmenting pro-survival signaling in the striatum and that disruption of p75 signaling at a pre-symptomatic age may exacerbate pathologic changes in these knock-in mouse models.
