Sonic hedgehog and TDP-43 participate in the spontaneous locomotor recovery in a mouse model of spinal motoneuron disease

Gulino R, Parenti R, Gulisano M (2017) Sonic hedgehog and TDP-43 participate in the spontaneous locomotor recovery in a mouse model of spinal motoneuron disease. J Funct Morphol Kinesiol 2:11.. doi: 10.3390/jfmk2020011

Summary: The authors used a mouse model of neurotoxic motoneuron depletion to study the role of Sonic hedgehog and TDP-43 in the compensatory changes occurring after the lesion. The injection of cholera toxin-B saporin (CTB-SAP; Cat. #IT-14) into the gastrocnemius muscle caused a partial motoneuron death accompanied by an impairment of locomotion.  Motor activity was significantly restored as soon as one month later.

Water-soluble chitosan inhibits nerve growth factor and attenuates allergic inflammation in mite allergen-induced allergic rhinitis

Chen PC, Hsieh MH, Kuo WS, Kao HF, Hsu CL, Wang JY (2017) Water-soluble chitosan inhibits nerve growth factor and attenuates allergic inflammation in mite allergen-induced allergic rhinitis. J Allergy Clin Immunol 140(4):1146-1149.e8. doi: 10.1016/j.jaci.2017.03.022 PMID: 28412394

Objective: To evaluate the effect of extensive water-soluble chitosan (EWSC) in Allergic rhinitis (AR).

Summary: The results suggest that NGF- and TH2-related cytokines created a positive feedback loop amplifying allergic inflammation in the upper-airway epithelial cells, and EWSC attenuated allergic inflammation and epithelial cell damage, by inhibiting NGF biosynthesis during allergic TH2 immune responses.

Sox2 is not required for melanomagenesis, melanoma growth and melanoma metastasis in vivo

Cesarini V, Guida E, Todaro F, Di Agostino S, Tassinari V, Nicolis S, Favaro R, Caporali S, Lacal PM, Botti E, Costanzo A, Rossi P, Jannini EA, Dolci S (2017) Sox2 is not required for melanomagenesis, melanoma growth and melanoma metastasis in vivo. Oncogene 36(31):4508-4515. doi: 10.1038/onc.2017.53 PMID: 28368402

Objective: To investigate whether Sox2 is required during the process of melanomagenesis, melanoma growth and metastasis and in the acquisition of resistance to BRAF inhibitors (BRAFi) treatments.

Summary: The findings exclude an oncogenic function for Sox2 during melanoma development and do not support a role for this transcription factor in the acquisition of resistance to BRAFi treatments.

Usage: Immunofluorescence on melanoma cell lines

A retinoraphe projection regulates serotonergic activity and looming-evoked defensive behaviour.

Huang L, Yuan T, Tan M, Xi Y, Hu Y, Tao Q, Zhao Z, Zheng J, Han Y, Xu F, Luo M, Sollars P, Pu M, Pickard G, So K, Ren C (2017) A retinoraphe projection regulates serotonergic activity and looming-evoked defensive behaviour. Nat Commun 8:14908. doi: 10.1038/ncomms14908 PMID: 28361990

Objective: To investigate how the dorsal raphe nucleus (DRN) and superior colliculus work in concert to extract and translate visual threats into defensive behavioural responses.

Summary: A dedicated population of RGCs signals rapidly approaching visual threats and their input to the DRN controls a serotonergic self-gating mechanism that regulates innate defensive responses.

Usage: Mice received bilateral intraocular injection (2 μg per eye) made between Streptavidin-Saporin and a biotinylated CTB antibody, or Anti-Melanopsin-SAP (2 μg per eye). For detection of melanopsin, retinas were incubated for 3 days at 4 °C with anti-melanopsin (1:600).
