Cross-talk between distinct receptors shapes itch behavior in the spinal cord.

Meng QT, Liu XY, Liu XT, Barry DM, Jin H, Yang Q, Sun Y, Wan L, Jin JH, Munanairi A, Kim R, Yin J, Tao A, Chen ZF (2018) Cross-talk between distinct receptors shapes itch behavior in the spinal cord. Neuron doi: 10.2139/ssrn.3249822

Summary: Consistently, Nppb-SAP ablated spinal Npr1 and Npr3 neurons and impaired histamine-, but not CQ-evoked, itch. Thus, the findings identify the role of BNP-NPRC signaling in modulation of histamine-evoked itch via NPRC-NMBR cross-talk independent of GRP-GRPR signaling. Our studies reveal distinct modes of action for bombesin-related peptides and NP in itch transmission.

Neuropharmacology of attention

Burk JA, Blumenthal SA, Maness EB (2018) Neuropharmacology of attention. Eur J Pharmacol 835:162-168. doi: 10.1016/j.ejphar.2018.08.008

Objective: This review focuses on the knowledge gained about neurotransmitter systems with respect to attentional processing, with emphasis on the functions mediated within the medial prefrontal cortex.

Summary: Future research needs to facilitate a better comprehension of the neural circuitry engaged by different subcategories of attention. Such information will be useful in allowing for more targeted treatments for conditions characterized by attentional deficits.

Usage: Several citations using 192-IgG-SAP;. The specific role of ACh in attentional function can be examined with lesions to cholinergic inputs to the mPFC, using the immunotoxin 192 immunoglobulin G (IgG) saporin. Increases in mPFC ACh efflux related to attention are no longer present following cholinergic lesions. These cholinergic lesions lead to severe deficits in performance on both sustained attention and 5CSRTT-related tasks.

Su Y, Liu Y, Behrens CR, Bidlingmaier S, Lee NK, Aggarwal R, Sherbenou DW, Burlingame AL, Hann BC, Simko JP, Premasekharan G, Paris PL, Shuman MA, Seo Y, Small EJ, Liu B (2018) Targeting CD46 for both adenocarcinoma and neuroendocrine prostate cancer. JCI Insight 3(17):e121497. doi: 10.1172/jci.insight.121497 PMID: 30185663

Objective: To investigate the suitability of a CD46 antibody as a metastatic castration-resistant prostate cancer (mCRPC) anti-tumor agent.

Summary: CD46 is an excellent candidate for antibody-based therapy development, which has potential to be applicable to both adenocarcinoma and neuroendocrine types of mCRPC that are resistant to current treatment.

Usage: Biotinylated UA20 IgG and Streptavidin-ZAP were mixed at a molar ratio of 1:1. The conjugate was used in cytotoxicity assays and shown to specifically kill the mCRPC cells.

Cell subpopulations overexpressing p75NTR have tumor-initiating properties in the c6 glioma cell line

Yang WH, Cheng CY, Chen MF, Wang TC (2018) Cell subpopulations overexpressing p75NTR have tumor-initiating properties in the c6 glioma cell line. 38(9):5183-5192. doi: 10.21873/anticanres.12841 PMID: 30194166

Objective: This study is focused on the possible role of p75NTR in glioma tumor initiation.

Summary: Cells with p75NTR overexpression demonstrated certain unique characteristics of tumor-initiating cells, such as neurosphere formation, high colony proliferation, and resistance to radio- and chemotherapy. With regard to the heterogeneous composition of glioma cells, p75NTR can be used as an alternative marker to identify a glioma subpopulation with tumor-initiating properties.

Usage: flow cytometry

Non-canonical Ret signaling augments p75-mediated cell death in developing sympathetic neurons

Donnelly CR, Gabreski NA, Suh EB, Chowdhury M, Pierchala BA (2018) Non-canonical Ret signaling augments p75-mediated cell death in developing sympathetic neurons. J Cell Biol 217:327. doi: 10.1083/jcb.201703120 PMID: 30018091

Summary: The authors investigated the interactions of pro-survival and pro-apoptotic receptors in Programmed Cell Death (PCD) using the sympathetic nervous system as a model. They demonstrate that Ret, a receptor tyrosine kinase required for the survival of many neuronal populations, is restricted to a subset of degenerating neurons that rapidly undergo apoptosis.
