Objective: To test the hypothesis that a neuronal population postsynaptic to nasal sensory neurons mediates sneezing.
Summary: Chemical activation of neuromedin B (NMB)-sensitive neurons elicits action potentials in caudal ventral respiratory group (cVRG) neurons and leads to sneezing behavior. This study delineates a peptidergic pathway mediating sneezing, providing molecular insights into the sneezing reflex arc.
Usage: Microinjection of NMB-saporin into the sneeze-evoking region abolished the sneezing responses to capsaicin and histamine.

Distinct roles of NMB and GRP in itch transmission.

Summary: To examine the role of NMBR+ dorsal horn neurons in itch and pain transmission, the authors treated mice with NMB-SAP (Cat. #IT-70; 2–3 μg, i.t.) at which no side effects were observed. NMB-SAP treated mice barely showed scratching behaviors, whereas control mice exhibited NMB-induced scratching with a rapid onset of scratching responses. These data demonstrate an important role of NMBR+ neurons in itch transmission. In contrast to notable deficits in itch transmission, NMB-SAP treated mice exhibited normal behavioral responses to acute mechanical stimuli, thermal stimuli and tail immersion. The authors also examined inflammatory pain behaviors of NMB-SAP treated mice and found normal nocifensive behaviors.

A nociceptive signaling role for neuromedin B.

Summary: Previous work suggests that neuromedin B (NMB) is involved in nociception. Direct injection of the peptide causes nociceptive sensitization, while NMB antagonists attenuate sensitization in reponse to nerve stimulation with mustard oil. Specific subsets of dorsal horn interneurons were eliminated by administering either 10 μg of the custom conjugate neuromedin B-SAP, 0.13 μg of SSP-SAP (Cat. #IT-11), or 1.3 μg of bombesin-SAP (Cat. #IT-40). Blank-SAP (Cat. #IT-21) was used as a control. The data indicate that NMB may be involved in the perception of thermal sensation, but not mechanical or pruritic sensation.
