Monoclonal antibody was obtained after BALB/c mouse immunization with the conjugate Nitroso-Cysteine-bovine serum albumin (BSA) proteins and hybridization of spleen cells with the myeloma cell line SP2/O/Ag14 (Clone number HY8E12). Ascites production was performed in BALB/c mice. The ascitic fluid was purified by ammonium sulfate precipitation.
This antibody targets conjugated NO-L-Cysteine. This antibody does not recognize free NO-L-Cysteine.
Antibody specificity was performed with an ELISA test by competition experiments with the following compounds: NO-L-Cysteine-G-BSA (1), L-Cysteine-G-BSA (1/200), NO-N-acetyl-cysteine-BSA (1/>50,000), N-acetyl-cysteine-BSA (1/>50,000).
Applications: ELISA, Immunohistochemistry (Glutaraldehyde and PFA fixation, see protocol), Immunocytochemistry. Optimal dilutions should be determined by each laboratory for each application. Suggested starting dilutions: 1/2000 – 1/10000 for ELISA, 1/1000 – 1/5000 for IHC
