Toolbox: Stable Transfected Cell Lines

CHOp75 Cells

192-IgG-SAP is the #1 rat p75-lesioning tool available for research use.  Our development of CHOp75 cells grew from the need for a stable, reliable cell line to use for Quality Control of each new lot of 192-IgG-SAP.  Now, these cells are available for use in laboratories worldwide.

CHOp75 cells are standard CHO (Chinese Hamster Ovary) cells that stably express the rat low affinity nerve growth factor, p75 (p75NTR). Expression of p75NTR in CHOp75 cells has been verified by flow cytometry with 192-IgG-Alexa488 (Cat. #AB-N43FLA) and cytotoxicity assay with 192-IgG-SAP (Cat. #IT-01).

CHO and CHOp75 cells were plated at 1000 cells/90 μl/well and incubated overnight. 192-IgG-SAP and saporin were added in 10 μl volumes, and the plates incubated for 72 hours. The cells were fixed with 10% TCA, then stained with 0.4% sulfarhodamine B/1% acetic acid. The plates were read at 564 nm. Data analysis done by Prism software (GraphPad, San Diego).

Several other transfected cell lines are undergoing final testing, including those expressing VGAT and the orexin-2 receptor.  These cell lines should be available soon.  Transfected cell lines are valuable tools that can be used for investigating the function of the transfected molecules.  They are also highly useful for screening research and therapeutic reagents that target the transfected gene product.  Contact us for more information about these cell lines or cell lines you would like to have developed?
